Single player is a story driven experience, complete with narrative and the puzzles are solvable purely with a single player / single portal gun.

However, coop requires the combined efforts of BOTH players (and both portal guns) to solve puzzles. They've been said to be very tricky at times, but very satisfying to solve.

Erm....

Single player is the story mode.

Co-op means you gang up with a pal to beat a level. Sort of like L4D where you play with friends to survive.

Single is like portal 1 style but co-op is 2 people working together on the same map to help eachother through the levels.

in single player it's just you. co-op requires you to have a friend or partner to complete the puzzles.

Co-op is an entirely different story, and involves seperate characters (two robots).

Single Player is the main story of the game. You play alone the whole campaign and finish the game like you normally would. Co-Op means you play a campaign with someone else. While usually it could be the main campaign, in Portal 2 they actually built a separate campaign just for Co-Op which expands on the story of Portal 2 :)

Single Player is way easier because there are only 2 portals but in co-op there will be 4 portals.
