You select the block that you want placed in your hotbar (row 1 of your inventory) by clicking it, scrolling to it with your mouse wheel or pressing its corresponding key in the numbers row of your keyboard. After that, you place your cursor over the tile that you want to be filled with your selected block and press left mouse button.
